Abstract
Objective: To determine the toxicity of the synthetic derivative 2-N-glycine-1, structurally related to 4-naphthoquinone lapachol, in Swiss male mice following oral administration of this compound. Methods: Ehrlich tumors were implanted into the subcutaneous tissue of the right axillary region of 8-week-old Swiss male mice, and the tumor cells were maintained in ascitic form. Treatments began on the third day after tumor cell implantation, and biometric assessments were performed daily until the eighth day. Microsteatosis and hepatic coagulative necrosis, Bowman's capsule thickening, and renal tubular necrosis were most severe with 1000 or 4000 μg/kg. Results: Overall, the most effective rate of tumor inhibition was observed in animals treated with 40 μg/kg of the compound, with a percentage reduction ranging from 90% to 96% compared to controls. Naphthoquinone did not exhibit a dose-dependent anticancer effect. However, significant dose-dependent histopathological alterations were observed, particularly in the liver, kidneys, heart, and spleen, as well as a decrease in hematocrit. Conclusion: The data demonstrate that while naphthoquinone promoted significant tumor reduction at a dose of 40 μg/kg, this was accompanied by marked dose-dependent toxicity in the liver, kidneys, and heart.
